About

Dihexa is a small synthetic peptide built from a two amino acid fragment of a natural blood pressure hormone. Chemists capped both ends with fatty acid chains to help it survive digestion and reach the brain. It was invented in a university lab as a possible Alzheimer's treatment. It became popular among people chasing memory and focus because its inventors claimed it built connections between brain cells at extraordinarily low concentrations. Three of the four papers behind that claim were retracted in 2025 after the university found the data had been faked. Dihexa has never been given to a person in any study. It is sold as an unregulated research chemical.

Dosing

Community practice is 5 to 10 mg per dose taken once or twice a week. Four independent first-hand reports land near 10 mg. Most people take it by mouth or under the tongue. Vendor dosing guides tell a different story and recommend daily use at up to 45 mg a day. No two of those guides agree with each other on dose, route, or units. Several contradict themselves on the same page. Nothing supports the higher figures. People commonly run it for a few weeks and then take a longer break.

Administration

  • Dihexa needs DMSO or another carrier because it does not dissolve in bacteriostatic water the way most peptides do
  • Vendor reconstitution guides that walk through bacteriostatic water math are copied from a generic peptide template and do not apply to this compound
  • The DMSO carrier irritates the skin of some people who apply it rather than swallow it

Half-life

No human study has ever measured how long dihexa lasts. Rat studies put the half-life near 12.7 days after a dose into a vein. That figure comes from the one paper that has carried an unresolved integrity flag since 2021. A separate number in the same paper, about 5.6 hours, describes how fast the peptide breaks down in a test tube rather than how long it lasts in an animal. The two are quoted interchangeably and they measure different things. The circulating claims of a two hour half-life and 50 percent oral absorption trace to vendor pages that admit elsewhere on themselves that no human data exists.

Storage

The powder keeps for about three years in a freezer and about two years in a fridge. Once it is dissolved in DMSO the solution keeps for about a month frozen. Reconstituted dihexa does not last the way a water-based peptide solution does.

Side effects

  • Headache is the most consistently reported effect and usually fades within days
  • Irritability that people tie to higher doses or to the off days of a cycle
  • Overstimulation or agitation as doses build up over several days
  • Changed sleep and vivid dreams reported in both directions since some people describe sleeping better
  • Skin irritation from the DMSO carrier rather than from the peptide itself

How it works

The mechanism people cite for dihexa no longer has evidence behind it. The claim was that it binds a growth factor called HGF and helps that growth factor switch on a receptor which builds connections between brain cells. The paper carrying that evidence was retracted in 2025 because the figures were fabricated. No outside laboratory has ever reproduced those measurements. Two independent groups did find that dihexa improves memory in mice and rats. One of them credited a completely different signaling pathway. A third independent group tested it in another disease model and found no effect at all. What dihexa does in a living brain remains unresolved.

Research quality

The evidence for dihexa is thin and partly discredited. Four papers from the lab that invented it were flagged for integrity problems in 2021. Three were retracted in 2025 after the university found figures had been faked and named two researchers as responsible. The fourth paper is the one that first made dihexa and supplies every dose, half-life, and brain-uptake number in circulation. It still carries the unresolved flag rather than a retraction or a clearance. The company that licensed the compound paid a four million dollar settlement in 2025 over the use of that data in a grant application. Dihexa itself has never been given to a person. A related molecule built from it did reach large human trials and missed both its main and its secondary goals in 2024. The claim that dihexa is millions of times stronger than a natural brain growth factor came from a press release rather than a published comparison.

Warnings

  • Dihexa has never been given to a person in any study so there is no human safety record of any kind
  • No animal study has ever looked for tumors, birth defects, or organ damage
  • It was claimed to act on a growth pathway that drives many cancers and no study has ever checked whether it promotes tumors
  • Avoid it in pregnancy and breastfeeding since no data exist in any species
  • The research-chemical supply has never had an independent purity audit so identity and dose accuracy cannot be verified
  • Vendor pages publish regulatory claims about dihexa that turn out to be false when checked against the actual government records
