About

Epithalon is a synthetic tetrapeptide with the sequence Ala-Glu-Asp-Gly. It is also written Epitalon. People take it for longevity and telomere claims. Its sequence was designed from the amino acid composition of a bovine pineal gland extract called Epithalamin. Epithalamin is a different substance and its human studies do not carry over. Epithalon is not an approved medicine anywhere and is not accepted for use in compounded medicines in the United States. Injectable material is sold as a research chemical with unverified strength and sterility.

Dosing

Sublingual dosing is the only route with an established amount. A trial gave 0.5 mg per day under the tongue for 20 days. The sublingual product sold in Europe delivers that same 0.5 mg per day for 30 days. Courses repeat after 4 to 6 months. A retinitis pigmentosa study used 5 mcg per eye by injection behind the eye for 10 days. A clinician performs that one rather than the person taking it. No injected dose has ever been established. The 5 to 10 mg figure that circulates for subcutaneous use is the package insert for a bovine pineal extract sold under a similar name with the route changed and the molecule swapped. First-hand injectable practice spans 50 mcg to 20 mg daily.

Administration

  • Vials are sold as either the free base or the acetate salt with the acetate delivering about 13% less peptide for the same labeled milligrams
  • A 10 mg vial covers an entire 20-day course at the published sublingual amount
  • Products sold as N-acetyl or amidated epithalon are different molecules and their per-spray amounts do not transfer

Half-life

No plasma half-life has been established for Epithalon in any species. FDA reviewers searching for pharmacokinetic data found none.

Side effects

  • Vivid or intense dreams from the first night onward that are reported more consistently than anything else
  • Daytime tiredness heavy enough that some people stop
  • Insomnia at higher doses in some people running opposite to the sedation others report
  • Stomach upset on the first day and flu-like aching around days 3 to 5
  • Facial swelling in one account that recurred across two vendors and two routes
  • Worsening wrinkles and loose skin in one account after months at 10 mg daily

How it works

Epithalon is the tetrapeptide Ala-Glu-Asp-Gly. Cell culture work reports that it raises hTERT expression, telomerase activity, and telomere length in normal human cells. An independent 2025 study reproduced that in normal fibroblast and mammary cells. The same study found something different in two breast cancer cell lines. It amplified an alternative telomere-lengthening pathway three to ten times there without raising telomerase enzyme activity. That study did not measure whether those cancer cells grew any faster. No human has ever had telomere length or telomerase activity measured while taking it. Sublingual dosing raised a urinary melatonin breakdown product and shifted the expression of three circadian genes in blood cells. Proposed binding to the telomerase gene promoter and to histone H1 comes from computer modeling rather than from measurement.

Research quality

Human evidence for the synthetic peptide amounts to two studies. A trial gave 0.5 mg sublingually for 20 days to 75 women and measured a urinary melatonin metabolite and three circadian genes rather than any sleep outcome. An uncontrolled series gave 162 people with retinitis pigmentosa 5 mcg per eye and reported vision improvements against historical comparison. No phase 2 or phase 3 trial exists. No toxicology program exists. No pharmacokinetic study exists. The telomere findings all come from cells in a dish. The founding 2003 report never published the concentrations it used. Rodent work reports fewer tumors and modest lifespan effects. The largest and longest of those studies found no change in overall tumor incidence. One rat study found its effect under a single lighting condition and not under two others. Almost all of that rodent work comes from the group that developed the compound. The long-running human mortality and geroprotection studies people cite belong to a bovine pineal extract rather than to this peptide.

Warnings

  • Most cancers become immortal by switching telomerase back on and this peptide is sold specifically to do that in adult tissue
  • Nobody has run the study that would show whether years of use changes cancer risk in either direction
  • People with a current or past cancer diagnosis have the least reason to accept an unmeasured risk of this kind
  • Purity certificates for this peptide test strength and cleanliness without testing for the clumping that regulators flag as the main immune risk
  • Clinics that sell it describe a compounding pharmacy source that regulators have been unable to find
  • Pregnancy, breastfeeding, and use in children have never been studied
