Humanin common uses, half-life, side effects and more

About

Humanin is a small peptide the body makes from a gene inside its mitochondria. Researchers found it while looking for something that protected brain cells from Alzheimer damage. People buy it for longevity, cognition, and metabolic goals. No registered or published trial has ever given it to a person. There is no human dose and no human safety record. Most of what is claimed for it was measured using HNG, a modified version that is far stronger in neuron tests. Neither form is approved as a medicine anywhere.

Dosing

Vendor pages give 0.5 to 2 mg under the skin. Others give 250 to 500 mcg or 2 to 5 mg two or three times weekly. All of those trace to unsigned dosing sites first published in 2026. Two of them print the same 0.5 to 2 mg figure five days apart. One calls it weekly and the other calls it daily. Fewer than ten people anywhere have publicly documented injecting it with a dose attached. The two clearest accounts used 0.4 to 1 mg and a single 200 mcg dose. Those forum posts date from 2017 and predate every dosing page. The animal doses behind the published results scale to roughly 11 to 57 mg in an adult. Marketed amounts sit well below that.

Administration

  • Check whether a vial holds native Humanin or the HNG analog because dosing pages give both the same milligram figure
  • A milligram injection is on the order of a thousand times the amount of Humanin the bloodstream normally carries
  • Doses are far below the animal amounts that produced the published results once those are scaled to human size

Half-life

Nobody has established how long Humanin lasts in a body. The one rodent study used HNG rather than the natural peptide. Mice cleared half of it in about 30 minutes after a dose into the abdomen. Rat readings were flat across the first four hours and lower at 24 hours with nothing sampled in between. No half-life can be read from that.

Side effects

  • Lasting facial swelling in one first-hand account after a single 200 mcg dose
  • Injection-site redness, swelling, or tenderness reported anecdotally
  • Glucose lowering is plausible from animal work though the human direction is unknown
  • No human adverse-event pattern exists because nobody has been studied

How it works

Humanin binds receptors on the cell surface and blocks the machinery that makes a cell kill itself. Two receptor candidates exist. One is a three-part complex built around gp130 and was worked out in neurons. The other is a formyl peptide receptor found on immune cells. Both feed into STAT3 and related survival signaling. Humanin also binds Bax and BimEL, two proteins that trigger cell death. That work used peptide already inside the cell rather than peptide arriving from outside. How an injected dose would reach those targets is unsettled. It binds a carrier protein called IGFBP-3 and that binding speeds clearance rather than slowing it. The receptor work used the natural peptide. Most animal results used the HNG analog instead.

Research quality

No registered or published trial has ever given Humanin to a person. The cell and rodent work is real and has been repeated by unconnected laboratories in Japan, France, the United States, Israel, and Argentina over 25 years. Roughly 70 to 85 percent of the animal work used HNG or another modified version rather than the natural peptide. Human research measures only the peptide people already make. Those studies contradict each other. Levels fall in Alzheimer disease in one and rise in another. The field has not settled whether they rise or fall with normal aging. Three papers on Humanin analogs in Alzheimer models have been retracted without a stated reason. The one mammalian lifespan test found no difference in lifespan.

Warnings

  • Injected Humanin made breast tumors grow faster and spread more in mice and the study authors warned against using it for other purposes
  • Two further laboratories independently found Humanin driving glioblastoma growth and chemotherapy resistance
  • The peptide works by stopping cells from dying on schedule and that is the ability a tumor needs to survive treatment
  • Anyone with a current or past cancer, or who is overdue for screening, is taking a risk nobody has measured
  • Vials sold as Humanin are often the HNG analog and dosing pages give both the same milligram amount despite calling HNG far stronger
  • Pregnancy, breastfeeding, and use in children have no safety data of any kind
